Of all the vast and varied Spider-Man cast members through the years, who has been your favorite and why?
For me, Mary Jane has always been that one redeeming force that always pulls Peter back from the edge after years and years of countless disappointments, beatings, and overall bad juju; she's the reason Spider-Man still swings across the city, his motivation after Ben and Gwen's deaths have been dulled by the passage of time. I think that's important, and she adds a lot to Peter's psychology; they're always each other's better halves when they need one.

I must say Mary Jane and Flash.
Mary Jane is a beautifully developed character and seeing her and Peter together is always a blast. I feel that they are indeed the perfect couple and Spider-Man wouldn't be who he is with his wife being there to support him. She's always there through the rough patches of his life and is always there to lift his spirits up when he's down. Everyone needs someone like that in their life and Mary Jane is that type for Peter.
Flash because for some reason, I loved the fact that they were rivals/enemies for YEARS and as time went on, they became really close friends. As years gone by, I tend to notice that people who you least expect to befriend end up being your closest friends and from little of what I've read of Spidey books, I would love to see when Flash would get out of his way to defend his friendship with Parker/Spider-Man after all those years of torturing the poor guy. :o

I have to say, I have always enjoyed the issues with Felicia Hardy, the Black Cat. She has had his back many times over the years. She can relate to him in some ways that even Mary Jane can't. I remember how she helped him when he had lost his powers. I think it was against Scorpion and Tarantula in the Powerless storyline. Also, she went out to avenge Spider-Man when he was defeated by Rhino in Sensational Spider-Man #34.
I would love to see Spider-Man and Black Cat back together if something happens to Mary Jane.

I second the Black Cat being a favorite, along with Betty Brant. If all you Mary Jane fans would read the older stuff (contained in Essential Spider-Man Volumes 2 thru 6) you would realize what a tramp she was. Gwen's body wasn't even cold when she made her move for Peter. I'm probably in the minority, but I can't wait until Brand New Day starts and the marriage is no more. Then Peter can get back to banging the Black Cat.

Black Cat. Hands down. She was exceptionally poorly written back when she was dating Spider-Man, turned from an interesting character into a ditzy air-head who would have spoiled Spider-Man's secret identity, just so the two could be broken up. I would actually love One More Day if it leads to Peter leaving MJ and being back with Felicia. She's proven over the years she has strong feelings for him, and would be an infinetely better partner for Spider-Man than a defensless celebrity target like Mary Plain.
Didn't Mary Jane leave Peter two or three times for going out there and saving lives instead of sitting at home with her? Even though she knew who he was and what he would be doing when she married him? Felicia understands Peter's dual identity better than MJ.

I'm a fan of Jameson and MJ.
As for Black Cat, she's a great read too. Also she wasnt air-headed, she was infatuated with Spiderman and didnt care for Peter Parker.

Officially, no she isn't. However that doesn't mean the comics she appeared in in that role never existed, nor does it mean that someone can't prefer that depiction of her to the rest of the supporting cast.
So Peter's wife, Mary Jane remains my favorite supporting character.
(Just as on the DC side I really prefer the pre-Crisis version of Huntress-- Bruce Wayne's daughter who hasn't been in continuity since 1986).

There are so many good supporting characters, but my favorites are MJ, Black Cat, and JJJ. They're all unique in their own aspects.
I just don't think Spidey could survive without MJ in some part of his life.
Top five in order -
Mary Jane Watson -Like most people, Mary Jane, his wife, no matter what Marvel is saying right now. :D ASM Issue 300, when she stripped for him when he was down man who didn't fall in love with MJ. :)
Flash Thompson - Big up to Flash Thompson, the bully turned best friend. Even when he was at his worst, he was always Spider-Man's #1 fan so you always knew that deep down he wasn't all bad. He also served in Viet Nam (I guess now you have to say he served overseas so he doesn't become really old all of a sudden ha ha) so he's a war hero too. Flash was really enjoyable in FNSM. After a bit of amnesia, him reverting to being a bully for a bit and then coming full circle, regaining his memories finding out that his best friend Peter Parker was Spider-Man was a huge turn around for the character. I wish of all people he didn't up and forget Peter was Spider-Man. Ditto for Aunt May too.
Aunt May - Speaking of, I have a soft spot for Aunt May. Even though she really should of remained dead after 400... If she would of died after OMD I wouldn't of had a problem with it actually. During the Lee/Ditko/Romita Sr. run she was really old anyway.
J.Jonah Jameson - You gotta love JJJ. That guy always cracks me up. :D
Human Torch - I'll toss in Flame Brain too, Spidey's best superhero bud.
Who's in your five LOL!

By far the best has to be JJJ. How can you not love J. Jonah? He is awesome and can make me laugh harder then Spider-Man can most of the time.

MJ would have to be my favorite, followed by Black Cat. That love triangle was gold.

JJJ, especially when we were able to delve deeper into the character and see why he actually hates Spider-Man and that it's not Spider-Man he hates, but himself.

Aunt May is the best supporting character. She is one tough old lady.
